I'll defer to Robert's experience, which is much more extensive than mine, but I would add this. Adventure Cycling recommends using a 40 spoke, 4x rear wheel for loaded touring. I would check out their web page or call them. I think there is a shop in Missoula MT where this non-profit touring organization is based that can build the wheels for you.

I also agree that you should be prepared to remove the free wheel to replace a drive side spoke. I have a "Hypercracker" that is very small and uses the chainstay to push against. All the spokes I have broken have been on the dirve side. There is also a temporary kevlar cord spoke that can be install without removing the free wheel. I don't remember the brand but I ordered mine from a local shop.
